After the cell lysate was transferred into a fresh tube, a 21-gauge needle was used to shear DNA and incubated on ice for 30 min. The lysate was centrifuged at 10,000g for 10 min to remove insoluble material. After that, it was incubated with protein A-Sepharose and subsequently precleared with Phex prebleed serum. Afterwards, the supernatant was collected and the anti-Phex antibody was added to it. This was performed overnight at 4degC.
In our experiments, we used membrane-associated Phex-WT or -3'M. We also used the supernatant from secreted hSTC-1 for viral stock production. To produce the recombinant baculovirus, we used Sf9 cells. These cells were diluted to one x 106 cells/ml and infected with recombinant baculoviruses at a multiplicity of five to ten. After 48 hours, we isolated the recombinant baculoviruses.
